چکیده

s.c. resulted in the death of all animals within 12 to 15 days. Treatment of infected mice with l,3-bis(2-chloroethyl)-lnitrosourea, 20 mg/kg, increased the range of death to 20 to 28 days with a significant number of long-term survivors (12%). The parameters utilized to determine the appearance of disease, induction of remission, and relapse following drug therapy were based on the bioassay of donor tissue at specified time intervals. When Bacillus Calmette-GuÃarin and Corynebacterium granulosum were used following 1 administration of l,3-bis(2-chloroethyl)-l-nitrosourea, 20 mg/kg, a high percentage of long-term survivors occurred in contrast to the groups of animals treated with immunostimulants or drug alone. The most important variable in these studies was the time in which immunostimulants were given following drug therapy. C. granulosum, when given 3 days after 1,3-bis(2-chlorethyl)1-nitrosourea therapy, resulted in 76% complete remissions, while B. Calmette-GuÃarin was most effective 12 days after drug treatment, yielding approximately 68% long-term sur vivors. The implications of these results to cancer therapy are discussed.

Determination of Monosomy 7 among In Vitro Generated CD1a Positive Cells in Patients with Juvenile Myelomonocytic Leukemia

Objective: To study whether the generated CD1a positive cells belong to the leukemic cells among patients with juvenile myelomonocytic leukemia. Materials and Methods: We used mononuclear cells from 3 patients with juvenile myelomonocytic leukemia, from which two had monosomy 7. The mononuclear cells from these patients were cultured in RPMI/10%FCS without adding exogeneous growth factors fo...

Establishment of modified retroviral vector targeting X-linked severe combined immunodeficiency.

Gene therapy targeting hematopoietic stem cells has been proposed as a potential therapy for numerous genetic disorders affecting hematopoiesis. Moloney murine leukemia retroviral vectors are now widely used for clinical gene transfer into hematopoietic progenitors and progeny. However, maintaining expression of therapeutic genes inserted via moloney murine leukemia virus (MoMLV)-based vectors ...

Combined VSV oncolytic virus and chemotherapy for squamous cell carcinoma.

OBJECTIVES Vesicular stomatitis virus (VSV) is a negative-strand ribonucleic acid (RNA) virus that replicates specifically in tumor cells and has oncolytic effects in a variety of malignant tumors. We previously demonstrated recombinant VSV vectors incorporating viral fusion protein (rVSV-F) and interleukin 12 (rVSV-IL12) to have significant antitumor effects against squamous cell carcinoma (SC...

Detection of abl/bcr Fusion Gene in Patients Affected by Chronic Myeloid Leukaemia by Dual-Colour Interphase Fluorescence in situ Hybridisation

Conventional cytogenetic is the standard technique for detection of Philadelphia (Ph) chromosome in chronic myeloid leukemia (CML). Evaluation of abelson murine leukemia/breakpoint cluster region (abl/bcr) fusion using dual-colour fluorescence in situ hybridization (D-FISH) is an alternative approach allowing rapid and reliable detection of the disease. We employed the technique of interphase D...
